Departs from the terminus at the corner of Victoria Street and The Esplanade and runs via Victoria Street, Kororoit Creek Road, Ferguson Street, Melbourne Road, Williamstown Road, Geelong Street, Geelong Road, Buckley Street, Victoria Street, Paisley Street, French Street, Irving Street, Leeds Street, Hopkins Street, Droop Street, Farnsworth Avenue, Fisher Parade, Langs Road, Chauvel Street, Monash Street, Progress Street, Charles Street, Epsom Road, Mirams Street, Francis Street, Rothwell Street, McCully Street, The Crescent, Ascot Vale Road and Mt Alexander Road to the terminus at the corner of Mt Alexander and Pascoe Vale Roads (Moonee Ponds Bus Terminus, Bay 2).

Route 472 from Moonee Ponds to Williamstown:

Departs from Bay 2 at the Moonee Ponds Bus Terminus at the corner of Mt Alexander and Pascoe Vale Roads and runs via the reverse of the route described above to Leeds Street, then via Paisley Street, Victoria Street and then via the reverse of the route described above to the terminus at the corner of Victoria Street and The Esplanade.
